At high power (40X), characteristic seminoma cells with variable cytoplasm, open (vs. clumped/hyperchromatic) nuclear chromatin pattern and prominent central nucleoli are seen. The cells do not impinge on each other as embryonal would.

Immunohistochemical stain for CD117/cKit shows membrane staining which confirms seminoma vs. embryonal, chorio and yolk sac testicular tumors. Other tumors such as GIST show this surface antigen staining, which is not specific for seminoma, but the stain differentiates this tumor from the other germ cell tumors.
